动物细胞实验室必备仪器进行简单介绍:1)CO2培养箱:CO2培养箱是细胞培养的必备仪器,它为细胞提供了一个体外培养的舒适环境,如适宜的温度、湿度、酸碱度、O2浓度等。细胞在体外培养时很容易受到各种细菌、微生物的感染,因此,CO2培养箱的灭菌和YJ能力就显得尤为重要;2)烘箱(干燥箱):用于细胞培养的一些器械、器皿必须烘干后才能使用,玻璃器皿等须干热消毒。干热消毒时,烘箱内温度一般要达到160℃以上,通常使用鼓风式烘箱。其优点是温度均匀、效果较好,缺点是升温过程较慢。升温时不能先升温后鼓风而应鼓风与升温同时开始,至100℃时,停止鼓风。需注意避免包裹器皿的纸或棉花烧焦,烧焦的碎屑可影响细胞的生长。消毒后不能立即打开箱门,以免骤冷导致玻璃器皿破裂,应等温度自然下降至100℃以下后可开门。3)超净工作台/生物安全柜:细胞培养对无菌环境要求很高,在细胞操作时,一般需要在100级空气质量条件下进行,因此超净工作台或生物安全柜就必须配备。4)液氮罐:为了保存细胞,特别是不易获得的突变型细胞或细胞株,要将细胞冻存。冻存的温度一般为液氮温度-196℃,因此,需要液氮罐。在此环境中,一般细胞可以保存十年具有活性。5)超低温冰箱:细胞的冻存过程需要一系列程序降温,一般4℃下存放30min,转放-20℃ 1 hour,再转入-70至-80℃过夜,之后才可以转移到液氮内(-196℃),这时就必须配备一台-86℃超低温冰箱。一般超低温冰箱保存细胞可达数月,对于不需要长期冻存的细胞,可暂时放于超低温冰箱保存。6)恒温水浴:从液氮或超低温冰箱中取出细胞冻存管需要立即放于37℃水浴中快速复苏之后才传代培养,因此,恒温水浴也是细胞实验室必备仪器。7离心机:细胞培养需要离心收集传代细胞,所以需要配置低速的常温离心机,对于后期细胞内容物的分离则需要高速冷冻离心机。除上述仪器外,移液器、高压灭菌锅、倒置显微镜、荧光显微镜也是细胞培养的必备仪器,发酵罐、转瓶机等也常常见于细胞培养实验室。

传代培养及其操作步骤:原代细胞培养成功以后,需要进行分离培养,否则细胞会因生存空间不足或密度过大,营养障碍,影响细胞生长。细胞由原培养瓶内分离稀释后传到新的培养瓶中培养的过程称之为传代培养。传代细胞允许培养的细胞扩增(形成细胞株),可以进行细胞克隆,易于保存,但可能丧失一些特殊的细胞和分化特征。传代细胞形成细胞株的利处在于提供了大量持久的实验材料,便于实验。【操作步骤】1)吸掉或倒掉培养瓶内旧培养液;2)向瓶内加入胰蛋白酶液和EDTA混合液少量。以能覆盖培养瓶底为宜;3)置37℃孵箱或室温(25℃温度)下进行消化,2~5min后把培养瓶放在倒置显微镜下进行观察,当发现胞质回缩、细胞间隙增大后,应立即中止消化;4)吸出消化液,向瓶内加入Hanks液少量,轻轻转动培养瓶,把残留消化液冲掉,然后再加培养液。如果仅使用胰蛋白酶消化,在吸除胰蛋白液后,可直接加入少量含血清的培养液,终止消化;5)使用弯头吸管,吸取瓶内培养液,按顺序反复轻轻吹打瓶壁细胞,使之从瓶壁脱离形成细胞悬液。吹打时动作要轻柔,以防用力过猛损伤细胞;6)用计数板计数后,分别接种于新的培养瓶中,置CO2培养箱中进行培养;7)细胞培养换液时间应根据细胞生长的状态和实验要求来确定。一般2~3d后应换一次生长液。待细胞铺满器皿底面,即可使用;也可继续传代扩大培养或换成维持液。【注意事项】1)掌握好细胞消化的时间,消化时间过短时,细胞不宜从瓶壁脱落,过长的消化会导致细胞脱落、损伤;2)掌握好消化浓度,当消化液浓度过高时,消化时间应缩短,过低时细胞消化时间相对延长。
