阴性染色产生的原因有： 　　（1）一抗和二抗浓度不合适或孵育条件不妥。 　　（2）荧光素提前衰退。荧光素质量不佳或操作过程中没有注意避光等防止荧光素衰退的事项。 　　（3）血清封闭时间过长。 　　（4）抗体稀释液PH值不合适，影响抗原抗体反应。 　　（5）组织切片不平、裂片或脱片很严重，易引起大块阴性着色。 　　（6）组织标本不新鲜或已经冷冻组织制成的切片中抗原易弥散，易引起本来表达的部位阴性染色或弱着色。 　　（7）荧光显微镜不会使用，激发波长选择错误。
