Low passage cells from pooled amniotic fluid specimens were cultured in 3 different media: Chang Amnio™, Amniomax™II and Amniomax™ C-100. The cells were seeded at 1X104 cells/well in triplicate in a 6-well-dish and grown without medium renewal for 6 days.

CHANG Amnio® is a complete, ready-to-use medium with an antibiotic developed for the culture of human amniotic fluid cells, chorionic villi sampling, and tissues for use in karyotyping and other antenatal testing. With an improved blend of basal media and serum, CHANG Amnio has been shown to yield higher growth than other commercially available media.
A variety of invasive and non-invasive techniques are used for successful prenatal diagnosis to monitor fetal gestation and detect fetal abnormalities. Chromosomes from Amniotic Fluid Cell (AFC), chorionic villi sampling (CVS) and tissues (POC) cultured in vitro have been used for more than 40 years to diagnose prenatal genetic abnormalities. A significant advance was reported by Chang et al. (PNAS, 1982) which subsequently led to the development of CHANG MEDIUM®, specifically designed for the culture of prenatal cells and tissues. Both the methods and the nutrient media used in the culture of these cells have evolved considerably over the years. To achieve optimum efficiency for diagnosis, a culture medium needs to promote rapid cell growth for a large number of colonies, fewer days to harvest, deliver a high mitotic index with sufficient metaphase cells and good chromosome morphology.
Chang Amnio™ is a new, high performing complete medium designed and optimized for providing analyzable metaphases for amniotic fluid, chorionic villi sampling and tissue (POC) cultures. It provides a reliable and consistent number of mitotic cells suitable for karyotyping for more accurate cytogenetic analysis. Chang Amnio™ is an excellent medium to choose when culturing prenatal cells.

CHANG Amnio羊水培养基使用注意

CHANG Amnio羊水培养基使用前准备
在37°C水浴锅边溶解边摇晃以便快速解冻;
CHANG Amnio®含抗生素,可根据需要自行添加抗生素。
分装
- 按照操作说明解冻;
- 在无菌的条件下分装到适用的体积并冷冻;
- 使用前将分装好的小包装37°C水浴即可。

Use of CHANG Amnio® for Primary Cultures: in situ Methodologies
原代培养:原位法
1. Centrifuge amniotic fluid at low speed to concentrate the cells.
2. Resuspend the cell pellet in a small volume of the patient’s own amniotic fluid.For example, aspirate the supernatant of 10 mL of spun amniotic fluid to 0.5mL above the cell pellet and resuspend. Add sufficient CHANG Amnio® to the concentrated cell suspension to allow for final plating volume of 0.5mL per cover slip (total of 4 coverslips) or 2mL per flaskette.
3. Incubate cultures undisturbed at 37°C 5%-8% CO2 atmosphere.
4. Flood cultures on day 2 by adding 2mL of CHANG Amnio®.
5. After 4 to 5 days, the cultures should be checked for growth. Cultures should be fed once growth has been observed. Feed cultures by removing all of the culture supernatant and replacing with 2mL fresh CHANG Amnio®. It is recommended that cultures be fed every 2 days thereafter.
6. Check cultures for growth on/or after day 5, and harvest when sufficient colonies are observed.
7. Best results are obtained when the cultures are fed with CHANG Amnio the day before the harvest.
Use of CHANG Amnio for Primary Cultures: Flask Methodologies
原代培养:培养瓶法
1. Centrifuge amniotic fluid at low speed to concentrate the cells.

4. Check for growth on day 5. Change medium with fresh CHANG Amnio and harvest if sufficient cell growth is observed.
5. Check cultures for growth and compley change medium every day thereafter until sufficient colonies are observed and are ready to harvest.
6. Best results are obtained when the cultures are fed with CHANG Amnio the day before the harvest.
Use of CHANG Amnio for Growing Passaged Amniotic Fluid Cells:

To passage the cells, treat the cultures with trypsin (or pronase, etc.) as you would normally do when cells are grown in conventional medium. However, protease treatment should be carefully monitored. Amniotic fluid cells grown in CHANG Amnio tend to be more sensitive to protease treatment than amniotic fluid cells grown in conventional medium. It may be necessary to modify your protocol to take this into account.The pH of the medium used to feed the cultures must be between 6.65–7.44 (i.e.the medium must be slightly yellowish salmon color).pH can easily be adjusted by placing the medium in a 5%-8% CO2 incubator with the cap slightly loosened for about 30 minutes.
Note: Calcium Oxalate crystals commonly form in CHANG Amnio. The presence of these crystals has not been shown to cause any detrimental effect on product performance.
