Description: Placenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family of growth factors. PlGF and VEGF share primary structural as well as limited amino acid sequence homology with the A and B chains of PDGF. All eight cysteine residues involved in intraand interchain disulfides are conserved among these growth factors. In their PDGF like regions, VEGF and PlGF also share approximately 53% amino acid sequence similarity. The gene for PlGF has been mapped to chromosome 14. As a result of alternative splicing, at least 3 PlGF mRNAs encoding monomeric PlGF precursors containing 149, 170 and 203 amino acid residues have been described. The expression of PlGF is not widespread, but has been detected in human umbilical vein endothelial cells, placenta, choriocarcinoma cell lines, and in renal cell carcinoma associated with angiogenesis. The PlGF proteins bind with high affinity to Flt-1, but not to Flk1/KDR. In addition to its action as a weak mitogen it is also a chemoattractant for monocytes and endothelial cells. PlGF is apparently a homodimer, but preparations of PlGF show some heterogeneity on SDS gels depending on the varying degrees of glycosylation. All dimeric forms possess similar biological activities. If PlGF is angiogenic in vivo is not clear yet. However, heterodimers between VEGF and PlGF are mitogenic for endothelial cells and have strong angiogenic activity in vivo (e.g. in the CAM assay or in the cornea pocket assay).
PlGF-3 is expressed exclusively in the placenta. It signals through the VEGFR-1/FLT1 receptor and stimulates endothelial cell proliferation and migration. PlGF-3 lacks heparin binding affinity. Recombinant human PlGF-3 is a 45.7 kDa disulfide-linked homodimeric protein of two 203 amino acid polypeptide chains.
