背景资料 This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis. Alternative promoter usage and alternative splicing result in dozens of transcript variants, but the full-length nature of many of these variants has not been determined.

单克隆抗体
抗原通常是由多个抗原决定簇组成的，由一种抗原决定簇刺激机体，由一个B淋巴细胞接受该抗原所产生的抗体称之为单克隆抗体（Monclone antibody）.单克隆抗体自问世以来，由于其独有的特征已迅速应用于医学很多领域。例如作为检验医学诊断的试剂。因为单克隆抗体有特异性强、纯度高、均一性好的优点，因此可以广泛应用于酶联免疫吸附试验、放射免疫分析、免疫组化和流式细胞仪等技术；单克隆抗体还可用于蛋白质的提纯。单克隆抗体是亲和层析中重要的配体。将单克隆抗体吸附在一个惰性的固相基质上，并制备成层析柱。当样品流经层析柱时，待分离的抗原可与固相的单克隆抗体发生特异性结合，其余成分不能与之结合。将层析柱充分洗脱后，改变洗脱液的离子强度或pH，欲分离的抗原与抗体解离，收集洗脱液便可得到欲纯化的抗原；除此之外，单克隆抗体还有望于参与肿瘤的导向ZL和放射免疫显像技术，将针对某一肿瘤抗原的单克隆抗体与化疗药物或放疗物质连接，利用单克隆抗体的导向作用，将药物或放疗物质携带至靶器官，直接杀伤靶细胞，称为肿瘤导向ZL。
单克隆抗体的优点
杂交瘤可以在体外存活并传代，只要不发生细胞株的基因突变，就可以不断地生产高特异性、高均一性的抗体。
可以用相对不纯的抗原，获得大量高度特异的、均一的抗体。
由于可能得到"无限量"的均一性抗体，所以适用于以标记抗体为特点的免疫学分析方法，如IRMA和ELISA等。
由于单克隆抗体的高特异性和单一生物学功能，可用于体内的放射免疫显像和免疫导向
