Description
G418 disulfate also known as geneticin, is an aminoglycoside antibiotic commonly used in plant and mammalian cell culture as a selectable marker. Plant cells which have been transformed with the npt II gene will be able to withstand G418 in the growth medium, making negative selection for successfully transformed plant cells possible.
TOKU-E offers three forms of G418: G418 disulfate (G001), G418 disulfate, EvoPure (G030), and G418 disulfate solution (G020). G418 disulfate and G418 disulfate, EvoPure? are soluble in aqueous solution. G418 disulfate solution is G418 dissolved in water at 50 mg/mL. EvoPure? products are purified single antibiotic fractions, most >99% pure.
Mechanism
G418 blocks polypeptide synthesis in eukaryotic cells by binding irreversibly to 80S ribosomes and therefore disrupting their proofreading capability.
Plant Biology Applications
Dong and McHughen (1992) compared the use of kanamycin and geneticin as a selection agent in the genetic transformation of flax by Agrobacterium mediated transformation. Experiments in sweet potato showed G418 to be a more reliable selection agent with a higher transformation efficiency compared to kanamycin.
