Description
Bacitracin is comprised of numerous antimicrobial peptide fractions. Notable fractions include A, A1, B, B1, B2, C, D, E, F, G, and X. Standard grade bacitracin (B002) is a mixture of these fractions. Bacitracin A is the most active and potent peptide fraction in bacitracin and is very similar to its bacitracin B counterpart.
TOKU-E offers seven forms of bacitracin: bacitracin (B002), bacitracin ultrapure (B014), bacitracin A EvoPure? (B015), bacitracin B1 EvoPure? (B016), bacitracin B2 EvoPure? (B017), bacitracin F EvoPure? (B021), and bacitracin Xa EvoPure? (B018). EvoPure? products are purified single antibiotic fractions, most >99% pure. High purity EvoPure? bacitracin products can be used to analyze the specifc effects of individual bacitracin fractions.
Mechanism
Bacitracin prevents phosphorylation of bactoprenol; a transport protein which carries peptidoglycan components outside the cell membrane. Without the active phosphorylated bactoprenol, peptidoglycan synthesis cannot be complete and the cell lyses. Two mechanisms of bacitracin resistance are understood. One mechanism relies on a protein called a BcrABC transporter which pumps bacitracin out after it has entered the cell. An alternative mechanism is due to another protein called BacA which provides the active phosphorylated bactoprenol from a different synthetic pathway.
Spectrum
Bacitracin primarily targets the cell wall in members of the gram positive bacteria including Streptococcus pyogenes and Staphylococcus aureus.
