Description
Carbenicillin disodium, ultrapure is a member of the carboxypenicillin β-lactam antibiotic family. TOKU-E offers two forms of carbenicillin: carbenicillin disodium (C004), and carbenicillin disodium, Ultrapure (C107). Both forms of carbenicillin are sparingly soluble in aqueous solution (0.45 mg/mL).
Mechanism
Carbenicillin interferes with PBP (penicillin binding protein) activity involved in the final phase of peptidoglycan synthesis. PBP’s are enzymes which catalyze a pentaglycine crosslink between alanine and lysine residues providing additional strength to the cell wall. Without a pentaglycine crosslink, the integrity of the cell wall is severely compromised and ultimately leads to cell lysis and death. Resistance to β-lactams is commonly due to cells containing plasmid encoded β-lactamases.
Spectrum
Unlike most β-lactams, carbenicillin disodium is limited to primarily gram negative bacteria including Pseudomonas aeruginosa and common enteric species.
Microbiology Applications
Carbenicillin is often preferred over ampicillin in selection protocols because of its greater stability and longevity resulting in less contamination by unwanted satellite colonies. In addition, carbenicillin disodium, Ultrapure is commonly used in clinical in vitro microbiological antimicrobial susceptibility tests (panels, discs, and MIC strips) against gram negative microbial isolates. Medical microbiologists use AST results to recommend antibiotic treatment options for infected patients. Representative MIC values include:Pseudomonas aeruginosa 64 μg/mL - >512 μg/mL

Carbenicillin has successfully been used in combination with Kanamycin in the transformation process of pear (cv. La France). Carbenicillin was succesfully selected as alternative bacterial growth regulator because of the low regeneration frequencies of this specific pear cultivar.
