The CD154 Detection Cocktail is designed for the easy identification and enumeration of activated antigen-specific CD4+ T cells by flow cytometry. To this end, cells are stimulated with the desired antigen in vitro to induce the expression of CD154 and simultaneously incubated with the CD154 Detection Cocktail. Besides a fluorochrome-conjugated CD154 antibody, the CD154 Detection Cocktail contains a CD40 blocking antibody to prevent the down-regulation of CD154 surface expression and a CD28 antibody to provide a costimulatory signal.
