氯氮平-N-氧化物 Clozapine N-oxide

5-羟色胺受体拮抗剂。Clozapine N-oxide is a major metabolite of Clozapine noted to decrease 5-HT2 serotonin receptor density in vitro. In studies with in vitro human liver microsomal preparations, Clozapine was observed to metabolize to N-Desmethyl Clozapine and N-oxide Clozapine. Clozapine N-oxide was noted to decrease 5-HT2 serotonin receptor density in rat primary cortical cells and human hippocampal SHS5Y5 neuroblastoma cells. As a method of identification, reversed-phase HPLC has been reported to be effective in identifying Clozapine and its two major metabolites.
