It is based upon a Sandwich assay principle and can be used to detect levels of Alpha-1-Antichymotrypsin as low as 0.938 nanomoles per milliliter.
About Alpha-1-Antichymotrypsin
Alpha-1-Antichymotrypsin is a plasma protease inhibitor and member of the serine protease inhibitor class. Polymorphisms in this protein appear to be tissue specific and influence protease targeting. Variations in this protein's sequence have been implicated in Alzheimer's disease, and deficiency of this protein has been associated with liver disease. Background
 
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- SERPINA3antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- SERPINA3 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the SERPINA3 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of SERPINA3 can be calculated.
