CD74抗体,Anti-CD74抗体产品介绍：CD74, also known as the MHC class II associated invariant chain (Ii), is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is produced in molar excess over MHC class II and some of this is expressed by an unknown pathway on the cell surface independent of, or in association with, MHC class II molecules. The half life of CD74 on the cell surface is only 3 to 4 min after which it is internalized. It has been proposed that cell surface CD74 binds The protein encoded by this gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF) which, when bound to the encoded protein, initiates survival pathways and cell proliferation. This protein also interacts with amyloid precursor protein (APP) and suppresses the production of amyloid beta (Abeta). Multiple alternatively spliced transcript variants encoding different isoforms have been identified.

固定的组织有可能破坏了原来组织的抗原结构，蛋白多肽发生交联，导致部分抗原表位封闭，结合位点减少，所以需要抗原修复，以暴露原来的抗原表位，达到充分显露抗原，以不出现明显脱片现象为佳。

抗原修复的几种常用方法（供参考）

1、 微波炉加热：

在微波炉里加热0.01枸橼酸钠缓冲液（pH6.0）至沸腾后将组织芯片放入，断电，间隔5-10分钟，反复1-2次。根据玻片情况可参考采用微波加热“3-5-3法”3分钟温火沸腾后静止5分钟，再温火沸腾3分钟即可。

适用的抗原：来源于人、大鼠、小鼠的组织标本；来源于其他动物种属的组织标本。

2、 沸热修复：

电炉或水浴锅加热0.01M枸橼酸钠缓冲液（pH6.0）至95℃左右，放入组织芯片加热10-15分钟。

3、 高压热修复：

在沸水中加入EDTA（pH8.0）或0.01m枸橼酸钠缓冲溶液（pH6.0）.盖上不锈钢锅盖，但不能锁定。将玻片置于金属染色架上，缓慢加压，使玻片在缓冲液中浸泡五分钟，然后将盖子锁定，小阀门将会升起来。10分钟后除去热源，置入凉水中，当小阀门沉下去后打开盖子。此方法适用于较难检测或核抗原的抗原修复。（骨及软骨组织的标本选择较温和的微波加热修复） 。

4、 酶消化方法：

常用0.1%胰蛋白酶和0.4%胃蛋白酶液。胰蛋白酶使用前预热37℃，消化时间约为5-30分钟。

适用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

抗体修FF式:

修复液：0.01M 枸橼酸（pH6.0）或者0.05M EDTA（pH8.0）

抗体修FF法：

方法1：沸水浴修复，将盛有修复液和玻片的烧杯置于沸水浴环境，保持外部沸腾状态15min，自然冷却至室温；

方法2：微波修复，将盛有修复液和玻片的烧杯置于微波炉中，高火5min，停火3min，中火5min，自然冷却至室温。
