Fibronectin fragment H-2572 inhibited fibronectin binding to the platelet membrane glycoprotein (GP) IIb/IIIa by interacting directly with this receptor, with an IC₅₀ of about 104 µM. HSRNSITLTNLT also inhibited the binding of fibrinogen to GPIIb/IIIa, as well as platelet aggregation. This indicates that this peptide derived from the cell binding domain of fibronectin comprises an RGD-independent GPIIb/IIIa binding site localized within the residues His¹³⁷⁷ and Ile¹³⁸².
