抗原的溶解方法：1：用移液器加 0.1ml 双蒸水到试管内，混匀，-20℃保存。若为其它规格（如：0.2ml）的冻干粉抗体，则加与标签上规格一致的（如：0.2ml）双蒸水溶解，混匀，-20℃保存。 2：用移液器加 0.05ml（规格的 50%）双蒸水到试管内，再加 0.05ml（规格的 50%）的甘油到试管内，混匀，-20℃保存；若为其它规格（如：0.2ml）的冻干粉抗体，则加标签上规格的 50%（如：0.1ml）双蒸水，再加标签上规格的 50%（如：0.1ml）甘油溶解，混匀，-20℃保存。

溶解要求：抗体溶解后，将其分装成 5-10 支（以 0.1ml 抗体为例，20ulx5 支或 10ulx10 支），-20℃保存，用一支取一支，避免反复冻溶。另：预试验可做几个梯度 1：100、1：200、1：400 稀释，如果背景高，还可再稀释，选一个佳的稀释比例，再正式做，效果。

抗体（antibody）指机体的免疫系统在抗原刺激下，由B淋巴细胞或记忆细胞增殖分化成的浆细胞所产生的、可与相应抗原发生特异性结合的免疫球蛋白。主要分布在血清中，也分布于组织液及外分泌液中。

保存条件 Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

荧光抗体在使用前应加以鉴定。鉴定指医学教丨育网整理标记包括效价及荧光素与蛋白质的结合比率。抗体效价可以用琼脂双扩散法进行滴定，效价大于1：16者较为理想。荧光素与蛋白质结合比率(F/P)的测定和计算的基本方法是：将制备的荧光抗体稀释至A2801≈1.0，分别测读A280 (蛋白质特异吸收峰)和标记荧光素的特异吸收峰，按公式计算。F/P值越高，说明抗体分子上结合的荧光素越多，反之则越少。一般用于固定标本的荧光抗体以F/P=1.5为宜，用于活细胞染色的以F/P=2.4为宜。抗体工作浓度的确定方法类似ELISA间接法中酶标抗体的滴定。将荧光抗体自1：4～1：256倍比稀释，对切片标本作荧光抗体染色。以能清晰显示特异荧光、且非特异染色弱的Z高稀释度为荧光抗体工作浓度。小量分装，-20℃冻存，这样就可放置3～4年。在4℃中一般也可存放1～2年。
