胰岛素抵抗PCR芯片 可检测参与
脂肪组织非胰岛素依赖型糖尿病(NIDDM)相关机制的84个关键基因的表达。消化食物时,胰岛素的释放激活胰岛素信号传导和细胞对葡萄糖的摄取,实现碳水化合物和脂质的合成和储存。胰岛素抵抗可能在患者病危期发生,特别是在长期高热量饮食之后,或者由其他风险因素造成。发生胰岛素抵抗的患者由于血糖原因,会并发多种疾病,包括非胰岛素依赖型糖尿病(NIDDM)、II型糖尿病,及肥胖等代谢综合征。胰岛素抵抗是肥胖和NIDDM之间的关键环节,可能由脂肪组织、胰岛、肝、骨骼肌之间的复杂调控网络失调造成。脂肪组织通过释放称为脂肪因子的激素调节食品的摄入量,并调控碳水化合物和脂质代谢。肥胖时,脂肪组织会发生慢性炎症,标志性的事件是炎性细胞因子的上调和白细胞浸润;而炎症反应的激活与胰岛素抵抗有关。芯片包括胰岛素和脂联素信号通路相关的脂肪基因、通常在NIDDM中失调的基因、先天免疫和炎症过程有关的基因,以及碳水化合物和脂质代谢过程中重要的酶和转运蛋白相关基因。通过实时定量PCR的方法,研究者即能够利用该芯片简单可靠地同时检测脂肪组织的胰岛素抵抗机制相关的基因表达。
