什么是Alvetex®?
Alvetex®是一种高度疏松的交联聚苯乙烯支架,形成的疏松孔洞直径约36-40μm,孔洞间的连通缝隙约12-14μm,并被截成200μm厚的薄片,该材料是化学惰性的,并且已经做成了适合常规细胞培养板的形式。
Alvetex®提供了一个非常合适的3D结构,细胞可以在这样更合适的环境里进行增殖、迁移、分化及执行各种生物学功能。细胞保持3D形态,并与相邻的细胞建立更紧密的相互联系。

Q: Are there any published papers concerning the use of alvetex®?
A: There are numerous peer-reviewed scientific articles available describing the development and application of alvetex®.
Q: Can I coat alvetex® with known proteins and reagents?
A: Yes. Most standard coating methods for cell culture plastic are compatible with alvetex®. For example, Collagen I and IV, Fibronectin, Laminin, Basement Membrane Extract have all been tested. See technical brochure showing example of collagen coated scaffold.
Q: Can I extract protein and nucleic acid from cells grown in alvetex®?
A: Yes. Cells can be disrupted and lysed within the scaffold using standard reagents for subsequent collection of total protein and nucleic acid.
Q: Can I remove alvetex® from the culture plate/insert?
A: Yes. alvetex® can easily be removed from the culture plate or well insert.
Q: Can I remove my cells from the scaffold?
A: Yes. Cells can be removed from the scaffold using a combination of trypsin and mild agitation. About 60-70% of cells can be removed in this way.
Q: Can I see cells using standard light microscopy?
A: alvetex® has been developed to enable 3D cell growth which it achieves very successfully. The visualisation of cells in any 3D system will always be challenging. More sophisticated methods such as confocal microscopy can be used to see cells cultured on alvetex® (see technical brochure). Depending on the cell density, it is possible to pick out individual cells growing within large voids.
Q: Can I use more than one scaffold within the same well?
A: It is possible to place more than one scaffold in a well or use the custom 6-well insert designed to house multiple scaffolds. This technology allows users for the first time the ability to study the interaction of two cultured cell types grown in 3D in an easy to handle system. Users can grow primary cells, or cells representative of a particular disease, and study their interaction with normal cells in these 3D models. This technology is designed to be very versatile with the user in mind.
Q: Does alvetex® come ready to use?
A: Like standard cell culture plastic-ware, alvetex® is ready to use from the moment it is unwrapped.
Q: Does alvetex® degrade?
A: alvetex® is inert and does not degrade during standard cell culture practice.
Q: Does alvetex® enable 3D cell growth?
A: Cells grown in alvetex® adopt a 3D geometry. Cells grow in contact with adjacent cells forming a thin 3D layer.
Q: Does the material auto fluoresce?
A: alvetex® does not auto fluoresce under the standard conditions we have tested for fluorescent microscopy.
Q: Has alvetex® been treated in any way?
A: alvetex® has been surface activated using the standard methods commonly used in preparing polystyrene for cell culture. alvetex® has also been sterilised using standard techniques.
Q: How do I seed cells onto alvetex®?
A: There are various ways of doing this. For example, users can seed cells in a small volume of medium (e.g. 200µl) centrall onto the scaffold, leave for 15min, and then carefully add the remaining culture medium. Alternatively, a cell suspension can be made using the full amount of culture medium and added directly to alvetex®. We recommend in the first instance, the user optimise the system for their purposes.
Q: How is alvetex® supplied?
A: alvetex® will be supplied packaged either in multi-well culture plates or in individual blister packed well inserts designed to fit all standard 6, 12 and 24 well plates.
Q: How much culture media do I use?
A: For any given culture vessel type, cultures grown in 3D contain more cells than their 2D counterparts. Accordingly, the metabolic demands on that media are generally higher in 3D. In a standard plate, the vessel should be filled with media to its maximum recommended level. To address this issue, a series of new devices has been developed to enable users to culture cells in 3D using alvetex® with larger volumes of culture media.
Q: How often do I need to change the culture medium?
A: This depends on the nature of the cells grown in 3D culture. Within a standard culture vessel, a 3D culture will require more frequent media changes compared to its 2D counterpart. However, this requires optimisation
for the cell type concerned. A series of new devices has been developed to enable users to culture cells in 3D using alvetex® with larger volumes of culture media. This equipment allows fewer media changes and maintains culture media quality.
Q: Is alvetex® available in a well insert?
A: alvetex® will eventually be available in well inserts as 6, 12 and 24-well formats.
Q: Is alvetex® compatible with other technologies?
A: alvetex® is a versatile material that can adapted for use in a broad variety of applications. We encourage investigators to explore its use in alternative and novel areas. Please enquire should you require advice about your particular application.
Q: Is alvetex® easy to use?
A: alvetex® is very straightforward to use. It has been developed specifically for routine 3D cell cultureand can be used alongside existing cell culture products. Full instructions are provided.
Q: Is alvetex® re-useable?
A: No, like any plastic consumable product for cell culture, it is not recommended to re-use alvetex®.
