 长期促销 人组织蛋白酶K（cath-K）ELISA Kit,人组织蛋白酶K  ，部分如下：
1. 标准品的稀释：本试剂盒提供原倍标准品一支，用户可按照下列图表在小试管中进行稀
释。
2. 加样：分别设空白孔（空白对照孔不加样品及酶标试剂，其余各步操作相同）、标准孔、
待测样品孔。在酶标包被板上标准品准确加样50μl，待测样品孔中先加样品稀释液40μl，
然后再加待测样品10μl（样品稀释度为5 倍）。加样将样品加于酶标板孔底部，尽
量不触及孔壁，轻轻晃动混匀。
3. 温育：用封板膜封板后置37℃温育30 分钟。
4. 配液：将30 倍浓缩洗涤液用蒸馏水30 倍稀释后备用
5. 洗涤：小心揭掉封板膜，弃去液体，甩干，每孔加满洗涤液，静置30 秒后弃去，如此
重复5 次，拍干。
6. 加酶：每孔加入酶标试剂50μl，空白孔除外。
7. 温育：操作同3。
8. 洗涤：操作同5。
9. 显色：每孔先加入显色剂A50μl，再加入显色剂B50μl，轻轻震荡混匀，37℃避光显色
10 分钟.
10. 终止：每孔加终止液50μl，终止反应（此时蓝色立转黄色）。
11. 测定：以空白空调零，450nm 波长依序测量各孔的吸光度（OD 值）。 测定应在加终止
液后15 分钟以内进行。

石蜡包埋组织切片3~4μm 厚度
1.烤片： 将待做切片置于切片架上，于60℃恒温烤箱中至少烤1 hr；
2.脱蜡： 切片放入盛有二甲苯的容器中脱蜡3 次（即二甲苯Ⅰ、Ⅱ、Ⅲ），每次
10 min；
3.水化： 切片经下行酒精水化，无水乙醇5min，95%乙醇2 次（每次2min），85%
乙醇2 min；75%乙醇2min，自来水冲洗，ddH2O 洗2×2min；
4.抗原修复： 根据抗体说明书推荐方法进行抗原修复，常采用高压、微波（温
度达到98~100℃）或酶消化修复法，室温自然冷却，自来水冲洗，ddH2O 洗2×
2min，TBS 洗涤（2×2min）（具体修FF法见附1）* 注：有些抗原勿需修复，
直接进入第5 步封闭。
5.封闭： 滴加试剂B，37℃湿盒孵育30 min；
6.加一抗：滴加用试剂C（即用型一抗），37℃湿盒孵育2 hr 或4℃一夜；
7.洗涤： TBS-T 洗涤（3×5 min）；
8.封闭： 滴加试剂B，37℃湿盒孵育10 min；
9.加二抗： 滴加用抗体稀释液稀释的生物素化二抗（试剂D），37℃湿盒中孵育
30 min；
10.洗涤： TBS-T 洗涤（3×5 min）；
11.封闭： 滴加试剂Tween 20，37℃湿盒孵育封闭20 min；
12.加HRP-SA： 滴加用试剂C 稀释的试剂E（1：50~200，终浓度5~20 nM），37
℃湿盒中孵育30 min；
13.洗涤： TBS-T 洗涤（3×5 min），TBS 洗涤（2×5 min）；
14.显色：应用DAB 溶液（试剂F）显色；
15.复染：自来水充分冲洗，复染，脱水，透明；
16.封片： 待组织标本干后，用试剂缓冲甘油封固剂封片；
17.观察成像： 显微镜下观察成像
