Anti-CD158e抗体,NK细胞YZ性受体3DL1抗体技术资料产品介绍:Killer cell immunoglobulin-like receptors (KIRs) are transmembrane glycoproteins expressed by natural killer cells and subsets of T cells. The KIR genes are polymorphic and highly homologous and they are found in a cluster on chromosome 19q13.4 within the 1 Mb leukocyte receptor complex (LRC). The gene content of the KIR gene cluster varies among haplotypes, although several "framework" genes are found in all haplotypes (KIR3DL3, KIR3DP1, KIR3DL4, KIR3DL2). The KIR proteins are classified by the number of extracellular immunoglobulin domains (2D or 3D) and by whether they have a long (L) or short (S) cytoplasmic domain. KIR proteins with the long cytoplasmic domain transduce inhibitory signals upon ligand binding via an immune tyrosine-based inhibitory motif (ITIM), while KIR proteins with the short cytoplasmic domain lack the ITIM motif and instead associate with the TYRO protein tyrosine kinase binding protein to transduce activating signals. The ligands for several KIR proteins are subsets of HLA class I molecules; thus, KIR proteins are thought to play an important role in regulation of the immune response.

NK细胞YZ性受体3DL1抗体保存条件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

修复液:0.01M 枸橼酸(pH6.0)或者0.05M EDTA(pH8.0)
抗体修FF法:
方法1:沸水浴修复,将盛有修复液和玻片的烧杯置于沸水浴环境,保持外部沸腾状态15min,自然冷却至室温;
方法2:微波修复,将盛有修复液和玻片的烧杯置于微波炉中,高火5min,停火3min,中火5min,自然冷却至室温。
常用试剂的配制
4、ELISA底物液(可溶性底物)
磷酸-柠檬酸缓冲液 pH5.
00.1mol/L 柠檬酸(21.014克/100ml):24.3ml
0.2mol/L Na2 HPO4(28.4克/1000ml):25.7ml
H2O:50ml
注:4°C存放备用
5、DAB底物液(不溶性底物,组化病理等)
0.05mol/L pH7.6 Tris-HCL
Tris:6.05克
HCL:(36%)3.15克(约2.7ml浓HCL)
H2O:至 1000ml
注:DAB为3´,3´二氧基联苯胺,不溶性底物,呈棕红色
6、DAB溶液配制:(组化及Western blot 显色)
称DAB 30mg,用0.05mol/L pH7.6 Trs-HCL缓冲液100ml溶解,如有沉淀,过滤后使用,一般新鲜配制使用;使用前加入1% H2O2 1ml至1.5ml,H2O2浓度为0.01%,混均后使用。
7、3′,3′,5′,5′,-四甲基联苯胺(TMB)溶液配制用二甲基甲酰胺(dimethylformamide,DMF)或无水乙醇,将TMB配成1%浓度,4°C保存半年以内使用;使用前,用pH5.0磷酸-柠檬酸底物液9.9ml加入0.1ml(1mg/TMB)1% TMB溶液中,再按每毫升TMB加入30% H2O2 1ul,混匀后立即使用。2N H2SO4终止后,450nm测定OD值。
8、1%离子琼脂胶配制液作免疫双相扩散,单相扩散及免疫电泳中使用。0.05U pH8.6 钠-HCL缓冲液。
