PGphArm Drug Screening The PGphArm system provides unique enabling tool for high throughput drug screening (efficacy and toxicity) process. It utilizes PepGel’s platform technology with superior properties and features for various cells culture or co-culture in 3D microenvironment as well as organ cells or organoid in 3D. PGphArm system is designed to mimic the natural extracellular matrix (ECM), a 3D network matrix composed of fibrous matrix proteins that provides structural integrity for cell survival, with a system that can be handled at room conditions. PGphArm system allows mimicking animal drug testing and accurately evaluating cell/organoid resistance to drugs.
The success of cancer therapeutics is mainly relying on the toxicity and efficacy screening tests of drugs. However, drug screening process is facing the challenges of low predictability in a timely manner. Animal model is still the most reliable model so far to answer the questions whether the drug is effective to kill the tumors without or the least damage to other organs. Animal model is not only expensive but also takes longer time for one cycle of testing.
Traditional cell based in-vitro tool is based on petri dish yielding not reliable results because the cell performance on a flat surface cannot reflect the real cell growth environment in vivo. The PepGel PGphArm system utilizes PepGel’s platform technology with superior properties and features for various cells culture or co-culture in 3D microenvironment as well as organ cells or organoid in 3D. PGphArm has the desirable mechanical and rheological properties for high throughput processing. PGphArm is the home for cells, biocompatible, injectable, so-gel reversible by slightly shearing force, easy for uploading of drugs and compounds at room condition or 37°C, compatible with cell media, superior microenvironment for human and animal cells culture. Drugs can be easily uploaded by two major methods: 1) Diffusion through the gel: drugs can be delivered to the cells living system (PGphArm gel system) by dispensing the drug solution into the cell culture media on top of the gel surface; 2) Pre-mix with PGphArm system: drugs can be pre-mixed with the PGphArm gel solution and the cell suspension before gelation.
