Mouse HLA class II histocomp*(代"a")tibility antigen gamma chain (CD74), is a single-pass type II membrane protein that in humans is encoded by the CD74 gene. It contains 1 thyroglobulin type-1 domain. CD74 Plays a critical role in MHC class II antigen processing by stabilizing peptide-free class II alpha/beta heterodimers in a complex soon after their synthesis and directing transport of the complex from the endoplasmic reticulum to comp*(代"a")rtments where peptide loading of class II takes place.

关于ULBP-1:
ULBP1, also known as RAET1I and NKG2DL1, is a member of the ULBP/RAET1 gene family. ULBP1 plays an important role in immune responses, especially in cancer and infectious diseases, and is well-known to bind to NKG2D together with at least ULBP 2 and 3. These proteins are distantly related to major histocomp*(代"a")tibility class I (MHC I) molecules, possessing the alpha 1 and alpha 2 Ig-like domains, but lacking the alpha 3 domain. Unlike MHC Class I, they have no capacity to bind peptide or interact with beta2-microglobulin. It can activate multiple signaling pathways in primary NK cells, gamma delta T cells, and CD8+ alpha beta T cells, resulting in the production of cytokines and chemokines.ULBP1 is expressed in wide range of tissues including heart, brain, lung, liver, bone marrow and some tumor cells, T-cells, B-cells, As an unconventional member of the MHC class I family, ULBP1 is able to interact with soluble CMV glycoprotein UL16 in CMV infected cells. The interaction with UL16 blocked the interaction with the NKG2D receptor, and thus might escape the immune surveillance. Furthermore, UL16 also causes ULBP1 to be retained in the ER and cis-Golgi apparatus so that it does not reach the cell surface. The ULBP1 regulation may have implications for development of new therapeutic strategies against cancer cells.

Tumor necrosis factor receptor superfamily member 9(TNFRSF9), also known as CD137 and 4-1BB, is an inducible T cell surface protein belonging to the tumor necrosis factor receptor superfamily. It is a single-pass type I membrane protein which contains 4 TNFR-Cys repeats. The human and mouse proteins share 60% amino acid sequence identity. CD137 is expressed by mesenchymal cells, including endothelial cells, chondrocytes, and cells of the central nervous system. CD137 is also broadly expressed by cells of the human immune system, is broadly expressed by cells of the human immune system, including activated CD8+ and CD4+ T cells, activated natural killer (NK) cells, follicular dendritic cells (FDCs) and monocytes. CD137 has diverse roles in the immune response, the one key function is to promote the survival of both T cells and dendritic cells by binding the cognate ligand CD137L (4-1BBL).
