关于Irisin/FNDC5:
Fibronectin type III domain-containing protein 5, the precursor of irisin, is a protein that is encoded by the FNDC5 gene.Human Irisin is synthesized as a 212 amino acid (aa) precursor encoding a type 1 transmembrane protein with a 121 aa extracellular domain (ECD), a 21 aa transmembrane domain, and a 39 aa cytoplasmic domain. The ECD of Irisin contains a fibronectin type III domain and multiple glycosylation sites. The ECD is proteolytically cleaved to release the 112 aa soluble Irisin hormone into circulation.Mature human, mouse share sequence identity.Irisin induces expression of peroxisome proliferatoractivated receptor γ coactivator 1α (PGC1α) and uncoupling protein1(UCP1), mitochondrialassociated metabolic proteins. Irisin induces the transition of white adipose tissue into more metabolically active beige adipose tissue.Irisin also regulates neuronal cell differentiation and neurite outgrowth in the brain and is involved in the differentiation of osteoblasts.

关于DKK-1:
This gene encodes a protein that is a member of the dickkopf family. It possesses two clusters of ten cysteine residues separated by a linker region. The protein highly expressed in thyroid, small intestine, stomach, liver, placenta, pancreas, uterus, abdominal cavity, bladder and skin. Weaker expression has been detected in colon and spleen. It antagonizes canonical Wnt signaling by inhibiting LRP5/6interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6.

CD64 (FcγRI), one of the Fc receptors for IgG, is a membrane glycoprotein that mediates endo*(代"c")ytosis, phagocytosis, antibody-dependent cellular cytotoxicity, cytokine release, and superoxide production. CD64 is also structurally distinct, containing an extracellular Ig-interactive region of three Ig-like domains in contrast to the two domains of the low affinity receptors FcγRII and FcγRIII. It is normally expressed on the surfaces of monocytes and macrophages.

Tumor Necrosis Factor Receptor Superfamily Member 21 (TNFRSF21) is a type I transmembrane receptor that includes four extracellular cysteine-rich motifs and a cytoplasmic death domain. DR6 is highly expressed in heart, brain, placenta, pancreas, lymph node, thymus and prostate. DR6 may activate NF-kappa-B and JNK to promote apoptosis and T-cell differentiation. In addition, DR6 binds with N-APP, which is released by the deprivation of Trophic-factor. It triggers caspase activation and degeneration of both neuronal cell bodies (via caspase-3) and axons (via caspase-6). DR6 is also expressed on the tumor cell lines and can be induced by TNF-α.
