Pull-down assays are useful for both confirming the existence of a protein–protein interaction predicted by other research techniques, and as an initial screening assay for identifying unknown protein–protein interactions. The minimum requirement is the availability of a purified and tagged protein (bait), which will be used to capture and "pull-down" a protein binding partner (prey). Principle and procedureGlutathione affinity is an efficient method for single-step purification of proteins carrying a GST (glutathione S-transferase) tag fusion. GST protein from various sources, both native and recombinantly expressed in E. coli and other host cells, can be purified by affinity chromatography on immobilized glutathione, followed by competitive elution with excess reduced glutathione. Glutathione HiCap resin represents a state-of-the-art matrix for affinity purification of GST and GST-fusion proteins via their strong and specific binding to the tripeptide ligand glutathione, which is stably immobilized on the bead support. The 26 kDa GST tag has a positive influence on protein solubility and high yields of highly pure proteins are easily obtained using a simple bind-wash-elute procedure (see figures "Glutathione affinity purification" , "Purification of GST-tagged protein using Glutathione HiCap Matrix", and "Glutathione HiCap Matrix delivers highly pure proteins"). The Glutathione HiCap Matrix is an excellent choice for column purification and scale-up (cartridges or custom-packed columns). Glutathione HiCap Cartridges are pre-filled with 1 ml or 5 ml Glutathione HiCap Matrix and are ready to use for purification of GST-tagged proteins using a syringe, peristaltic pump, or automated liquid chromatography systems (such as ÄKTA, ÄKTAdesign, or BioLogic workstation). The highly physically stable bead support (WorkBeadsTM) is made from agarose, providing reliable purification for a wide range of proteins and under a variety of conditions. The matrix combines high resolution with a high binding capacity and allows high flow rates (see figures "Superior binding capacity (batch format)", "Superior binding capacity (gravity flow)", and "Dynamic binding capacity of Glutathione HiCap Matrix"). Glutathione HiCap Matrix is suitable for batch purification, gravity-assisted chromatography, and packed-bed column chromatography (cartridges or custom-packed columns), as well as for high-throughput applications in vacuum- or centrifugation-based protocols. Glutathione HiCap Matrix is also well suited for GST pull-down applications, where the beads are subjected to repeated high g-force centrifugation steps. Monoclonal GST-tag antibodies are used to detect GST-tagged proteins in immunodetection procedures with high sensitivity and specificity (see figure "Sensitivity and specificity of GST-tag Antibody").
