Glucagon is a secreted protein and belongs to the glucagon family. Glucagon can be cleved into 8 chains, playing an important role in initiating and maintaining hyperglycemic conditions in diabetes. Glucagon can regulates blood glucose by decreasing glycolysis and increasing gluconeogenesis. In addition, Glucagon is involved in initiating and maintaining hyperglycemic conditions in diabetes. Glucagon release is stimulated by hypoglycemia and inhibited by hyperglycemia, insulin, and somatostatin. In the glucagon antagonist, His-53 and Phe-58 are missing. This antagonist has been successfully utilized to reduce glucose concentration in vivo.

关于B7-H1/PD-L1：
CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumor growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.

  本酶的常见切割位点位于Ile-Glu/Asp-Gly-Arg 序列中的精*酸残基之后。有时由于蛋白质底物构象的变化，Factor Xa蛋白酶也可能于其它碱性*基酸残基处发生切割。在这些已经测序的*基酸残基中，最常见的次级识别位点是Gly-Arg序列。在 E.coli 中不稳定的蛋白质和被Factor Xa在次级识别位点切割的蛋白质之间似乎存在某种相关性；这似乎表明这些蛋白是处于部分未折叠的状态（Walker, I., Riggs, P.，该实验结果未发表）。Factor Xa 蛋白酶不切割识别序列后是脯*酸或精*酸的识别位点。

来源：Factor Xa蛋白酶是从牛血浆纯化得到，经RusseII蛇毒中的活化酶活化处理。

活性检测条件：50μl反应体系中，20mM Tris-HCl,100mM NaCl,2mM CaCl2（pH8.0 储存于 25℃）, 50μg MBP融合蛋白测试底物。6小时内，超过95%的融合蛋白能够被Factor Xa完全切割。

活性定义：在6小时或更短时间内，1μg的 Factor Xa能够完全切割95%的50μg测试底物。

储存液：20mM HEPES（pH 8.0储存于 4℃），500mM NaCl，2mM CaCl2，50%甘油。

储存条件：-20℃。
