BLMH / Bleomycin Hydrolase抗体 Background
Bleomycin hydrolase, also known as BLM hydrolase and BLMH, is cytoplasm protein which belongs to thepeptidase C1 family. BLM hydrolase / BLMH is a cytoplasmic cysteine peptidase that is highly conserved through evolution. The only known activity of the enzyme is metabolic inactivation of the glycopeptide bleomycin (BLM), an essential component of combination chemotherapy regimens for cancer. It catalyzes the inactivation of the antitumor drug BLM (a glycopeptide) by hydrolyzing the carboxamide bond of its B-aminoalaninamide moiety thus protecting normal and malignant cells from BLM toxicity. BLM hydrolase / BLMH contains the signature active site residues of the cysteine protease papain superfamily. BLM hydrolase / BLMH inactivates bleomycin B2 (a cytotoxic glycometallopeptide) by hydrolysis of a carboxyamide bond of beta-aminoalanine, but also shows general aminopeptidase activity. The specificity varies somewhat with source, but amino acid arylamides of Met, Leu and Ala are preferred. BLM-induced death and has an important role in neonatal survival and in maintaining epidermal integrity. Mice lacking BLMH exhibited variably penetrant tail dermatitis that resembled rodent ringtail. The histopathology of the tail dermatitis was similar to skin lesions in humans with pellagra, necrolytic migratory erythema, and acrodermatitis enteropathica.
