关于IL-13：
Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13Rα1 with low affinity and triggers association of IL4Rα, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13Rα2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

关于LAIR2：
Leukocyte-Associated Immunoglobulin-Like Receptor 2 (LAIR2) is a secreted, 131 amino acid protein that contains one Ig-like C2 type domain, making it a member of the Ig superfamily. When comp*(代"a")red to LAIR-1, its transmembrane counterpart, it shares 83% amino acid identity across the signal sequence and extracellular domains; although one is secreted and one is membrane-bound, the two LAIR proteins are thought to have arisen from a common gene ancestor and appear to share similar adhesion profiles. This suggests that LAIR-2 may compete with LAIR-1 for ligand binding. A 114 amino acid alternate splice form of LAIR-2 is truncated at the C terminus, but retains the entire Ig domain. The expression profile of these splice forms, and the presence of orthologs in other species, have not been reported.

  本制品是将PCR反应所需的Pfu酶、dNTP、MgCl2、反应缓冲液以及优化剂以及稳定剂，预先配制成2倍浓度的混合物。2×Pfu PCR MasterMix专为扩增克隆DNA片段优化，高保真，错误率仅为1.3×10-6。使用 时只需在加入模板和引物并稀释到1倍浓度即可进行PCR反应，非常方便。用于要求保真度比较高的PCR反应，包括克隆PCR、DNA片段拼接、引入突变、全基因合成等。

使用建议：
Pfu DNA聚合酶产生的PCR产物为平滑末端，无3´端"A"突出，其PCR产物的克隆有几种方案。
1、PCR前引物进行5´端加磷修饰或将PCR产物磷酸化处理后再直接克隆于平滑末端的载体中（参见实验方案）。
2、将产物3´末端加A后再与T载体连接（参见实验方案）。
3、引物中引入15bp与载体同源序列，利用BalbRec PCR产物一步定向无缝克隆试剂盒（Cat# JN0001）直接连接到目的载体。
由于Pfu DNA聚合酶的校对活性可引起引物从3´端被部分降解。因此在设计引物时应适当增加引物的长度，理想的引物长度为20-30碱基。另外为了减少由3´→5´外切酶活性引起的引物降解，尽量在冰上配制反应体系，并加入Pfu酶。

应用实例：

图例一）50μl 扩增体系中，分别以50ng～0.05ng人基因组DNA为模板，可对特定基因片段（380bp）进行很好的扩增。
