关于CAR:
Coxsackievirus and Adenovirus Receptor (CAR) belongs to the CTX family of the Ig superfamily. CXADR is a type I transmembrane glycoprotein and expressed in pancreas, brain, heart, small intestine, testis, prostate. It is a receptor that mediates gene transfer and also act as an adhesion molecule within junctional complexes, notably between epithelial cells lining body cavities and within myocardial intercalated discs. CXADR contains an extracellular domain, a transmembrane helix and a C-terminal intracellular domain. The C-terminal interacts with few cytoplasmic junctional proteins, microtubules and the actin cytoskeleton.

关于FcγRIIB:
FcγRIIB is a low affinity receptor that recognizes the Fc portion of IgG. The human CD32 group consists of FcγRIIA, FcγRIIB, and FcγRIIC proteins that share 94-99% sequence identity in their extracellular domains but differ substantially in their transmembrane and cytoplasmic domains. FcγRII protein is expressed on cells of both myeloid and lymphoid lineages as well as on cells of non-hematopoietic origin. FcγRIIB has an intrinsic cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM) and delivers an inhibitory signal upon ligand binding. Ligation of FcγRIIB on B cells down-regulates antibody production and in some circumstances may promote apoptosis. Co-ligation of FcγRIIB on dendritic cells inhibits maturation and blocks cell activation. FcγRIIB may also be a target for monoclonal antibody therapy for malignancies. FcγRIIB plays an important negative-regulating role through modulating the signals from activation receptors.

Programmed Death-1 (PD-1), firstly cloned from mouse T cell hybridoma 2B4.11, is one member of CD28/CTLA-4 superfamily. PD-1 belongs to type I transmembrane protein and acts as an important immunosuppressive molecule. The cytoplamsic tail of PD-1 contains two structural motifs, an immunoreceptor tyrosine-based inhibitory motif (ITIM) and an immunoreceptor tyrosine-based switch motif (ITSM) formed by two tyrosine residues which make the difference in PD-1 signal mediating.Mouse PD-1 is expressed in thymus and shares about 69% aa sequence identity with human PD-1. Recently, programmed death-1 (PD-1) with its ligands, programmed death ligand B7H1 (PD-L1) and B7DC (PD-L2), was found to regulate T-cell activation and tolerance, upon ligand binding, inhibiting T-cell effector functions in an antigen-specific manner.PD-1 gene knocked out mice would induce some autoimmune diseases, which suggests that PD-1 acts as a co-inhibitory molecule actively participating in maintaining peripheral tolerance. Thus, PD-1 may be a useful target for the immunologic therapy of carcinoma,infection,autoimmune diseases as well as organ transplantation.

Cytoplasmic Protein NCK1 (NCK1) is a cytoplasmic protein that contains one SH2 domain and three SH3 domains. NCK1 is a member of the adapter family, which associates with tyrosine-phosphorylated growth factor receptors, such as KDR and PDGFRB, or their cellular substrates. NCK1 maintains low levels of EIF2S1 phosphorylation by promoting its dephosphorylation by PP1. NCK1 plays a role in the DNA damage response, but not in the detection of the damage by ATM/ATR. It is also involved in transducing signals from receptor tyrosine kinases to downstream signal recipients, such as ELK1-dependent transcriptional activation in response to activated Ras signaling.
