Introduction Alpha-Macroglobulin is a major serum protein with diverse functions, including inhibition of protease activity and binding of growth factors, cytokines, and disease factors (1). Increased serum α-macroglobulin has been suggested to be associated with multiple sclerosis (MS) (2), glomerular disease (3), and with liver diseases (4). Rat α-macroglobulin (Rat AMG) is believed to be a functional homologue of human α2-macroglobulin (Human α2M).
Principle of the Assay The AssayMax Rat α-Macroglobulin ELISA kit is designed for detection of rat α-macroglobulin in rat plasma, serum, urine, and cell culture supernatants. This assay employs a quantitative sandwich enzyme immunoassay technique that measures α-macroglobulin in less than 4 hours. A polyclonal antibody specific for α-macroglobulin has been pre-coated onto a microplate. Rat α-macroglobulin in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for α- macroglobulin, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
