The purpose is to decrease the signal caused by endogenous avidin, biotin, or biotin-binding proteins when immunostaining specimens with detection systems that rely on avidin-biotin or steptavidin-biotin chemistry. Suggested Working Dilution |  | The optimal working dilution should be determined for each specific assay condition. Blocking Procedure: Avidin/Biotin Blocking System (SIGNET)For paraffin-embedded tissues: (To be performed before primary antibody incubation and after antigen retrieval). 1. Incubate the Avidin Solution for 10-15 minutes at RT. 2. Rinse with PBS or TBS buffer 3-4 times. 3. Incubate with the Biotin Solution for 10-15 minutes at RT. 4. Rinse with PBS or TBS Buffer 3-4 times. |
